The 'Lock Keyframes' setting is controlled by clicking the button at the far right of the timetrack at the bottom of the canvas window.  The icon has a picture of a padlock on it, with arrows pointing left and right, and will only be visible if your canvas has a non-zero end time.  It will only be active if you're not currently using a tool which disables the timetrack, such as the bline tool or the draw tool.
